Abstract Context Withania coagulans (Stocks) Dunal fruits are used in the therapeutics of several ailments due to possessing potent phytoconstituents which is also traditionally for curing diabetes. Objective The present study was assessing amelioration potential phytochemicals an ethanol fruit extract W. HOMA (Homeostatic model assessment) indices and pancreatic endocrinal tissues by inhibition DPP-4 antioxidants activities. Material methods identification test performed LCMS. Further, assessments in-vitro, in-vivo in-silico were achieved following standard methods. In-vivo studies conducted on type-2 diabetic rats. Results chosen inhibited activity 63.2% vitro assay as well significantly inhibit serum levels. Accordingly, administration resulted a significant ( P ≤ 0.001) alterations lipid profile, antioxidant levels, indices. Moreover, (islet Langerhans) appeared have restoration normal histoarchitecture evidenced increased cellular mass. Molecular docking (Protein-ligands) identified with (target enzyme) shown incredibly low binding energy (Kcal/mol) required ideal interactions. ADMET analysis pharmacokinetics indicated profile per Lipinski laws. Conclusion It can be concluded that result improved glucose homeostasis
